چکیده

UNLABELLED This feasibility study was designed to assess the ability to measure mitochondrial DNA (mtDNA) in cerebrospinal fluid (CSF) cells that contributed to minimal disease/persistent or residual disease (MD/PRD) from children with acute lymphoblastic leukemia (ALL). Increase in mtDNA copies in cancer cells has been suggested to play a role in MD/PRD. CSF as well as blood specimens from 6 children were assayed for MD/PRD and mtDNA copy numbers by quantitative real-time polymerase chain reaction. Of 7 MD/PRD-positive specimens, 6 had increased mtDNA copy numbers; while 11 MD/PRD-negative specimens had no increase in mtDNA copy numbers, p < 0.003. This is the first proof-of-concept study to measure mtDNA copy numbers in MD/PRD-positive CSF specimens from children with ALL. Increase of mtDNA copy numbers in MD/PRD childhood ALL cells and its significance as a mechanism for recurrence requires further investigation. KEYWORDS Minimal residual disease; Acute lymphoblastic leukemia; Central nervous system; Cerebrospinal fluid; Mitochondria.

The role of supine and prone positions on central nervous system involvement in patients with acute lymphoblastic leukemia

Background: Acute lymphoblastic leukemia (ALL) affects both children and adults, with a peak incidence between the ages of 2 and 5 years. ALL cells sometimes penetrate the central nervous system (CNS) and patients with CNS diseases at initial presentation have been reported to experience a significantly greater risk of treatment failure compared with CNS negative patients. This study hypothesiz...

Influences of Genetic Abnormality on the Risk of Acute Lymphoblastic Leukemia

Recent studies have provided evidence that common genetic variations could account for a proportion of leukemia in adult or children. To evaluate the contribution of candidate gene association studies to the understanding of genetic susceptibility to acute lymphoblastic leukemia we conducted a systematic review from published studies. The polymorphisms of genes encoding carcinogen-metabolizi...

The relation between end of induction minimal residual disease and different risk factors in patients with acute lymphoblastic leukemia

Background: Malignant disorder with B or T stem cell basis leads to development and continuation of acute lymphoblastic leukemia (ALL) due to aggregation of blast cells in bone marrow. The environmental, genetic, and demographic factors may influence the disease relapse. The objective of this study was to assess the relation between end of induction minimal residual disease and different risk f...

Epigenetic effects of decitabine on acute lymphoblastic and acute promyelocytic leukemia cells

Background: Decitabine (5-aza-2'-deoxycytidine, DAC) is a deoxycytidine analog currently used as an effective drug against myelodysplastic syndromes and acute myeloid leukemia. Although various studies have pointed out the epigenetic effects of this drug, its epigenetic mechanisms in different leukemic cell lines are not specified. In this lab trial study, possible epigenetic effects of decitab...
